TNFα stabilizes RARα protein by activating NF-κB. (A) NB4 cells were treated with 10 ng/mL TNFα for the times indicated, followed by western blotting for proteins as indicated. (B) NB4 cells were treated with 10 ng/mL TNFα for 0.5 h, followed by immunofluorescent staining using anti-p65 antibodies. (C) NB4 cells with NC or sh-p65 infection were treated with 10 ng/mL TNFα for 12 h, followed by western blot analysis for proteins as indicated. (D) NB4/GFP and NB4/GFP-MAD cells were treated with 10 ng/mL TNFα for 0.5 h, and then the intracellular localization of p65 was analyzed by immunofluorescence. (E) NB4/GFP and NB4/GFP-MAD cells were treated with 10 ng/mL TNFα for 12 h. The cell lysates were western blotted for the indicated proteins. All experiments were repeated three times and gave consistent results.
