Morphology of single hematopoietic and AML stem cell clones. Single cells, either hematopoietic stem cells (HSCs) or ASCs, were placed in separate wells within 96-well plates and cultured in serum-free medium containing stem cell factor, Flt-3, thrombopoietin, and granulocyte colony-stimulating factor. After 5 days of culture, each well was examined using an inverted microscope to determine growth and plating efficiency of the single stem cell. (A) Normal HSCs obtained from adult bone marrow showed variable degree of proliferative potentials from Grade 1 to 4. (B) Almost all of HSCs obtained from cord blood showed high proliferative properties at Grade 4. (C) The plating efficiency of single ASCs remained at a remarkable low level at Grade 1.
