Cyclin D1 expression impairs the IRE1 and PERKS pro-apoptotic axes of the UPR after bortezomib treatment. 8226- and LP1-derived clones were treated with vehicle (Cont) or treated with 5 or 10 nM bortezomib for 24 h. The cells were then harvested, and whole-cell proteins were prepared and analyzed by SDS-PAGE and in Immunoblotting with anti-XBP1 (A) or EIF2α (B) Abs. The anti-β-actin Ab was used as a control for charge and transfer. The experiments were carried out two times; one is presented. The spliced and active form of XBP1 is denoted “s”, the unspliced form is denoted “u”; the cleaved form of eIF2α is indicated by an arrow.
